General description

Molecular cytogenetics allows for the study of unexplored systems, by bridging the gap between molecular biology (a few hundred Kb), and high resolution cytogenetics (approx. 2-5 Mb).  Molecular cytogenetics which is a relatively new technology is mainly based on fluorescence in-situ hybridization (FISH) applied to meiotic and mitotic chromosome preparations and DNA fibres, and has achieved a real breakthrough in cytogenetics.

Know-how, expertise available

The molecular cytogenetics core facility provides know-how and expertise in fluorescence in-situ hybridization (FISH) applied to meiotic and mitotic chromosome preparations:

  • Mapping DNA sequences on chromosomes (FISH),
  • Genomic in-situ hybridization (GISH) for the differentiation of parental chromosomes in interspecific hybrid and polyploid structures.
  • Physical mapping and organization of BAC clones to chromosomes (BAC-FISH) applied to meiotic and mitotic chromosome preparations (pachytene stage) for a higher resolution,
  • BAC-Fiber-FISH technology applied to combed DNA fibres for sequence mapping and determination of physical length in kilobases.

  • Cytogenetic characterization of plant material from interspecific hybrids and polyploid species for chromosome identification in each species or introgression,
  • Genomic structural evolution and behaviour in polyploid species,
  • Characterization of chromosomal translocations based on genetic and physical mapping (cytogenetic mapping),
  • Physical mapping of meiotic chromosomes (pachytene stage) and DNA fibres allowing for minute characterization (< 1 Kb) of the locus of interest and detection of chromosomal rearrangements.
